PURPOSE: A method for establishing a cell surface expression system using an autotransporter is provided to enable secretion of target proteins. CONSTITUTION: A surface expression vector contains a signal sequence, a passenger domain, a linker site, and a cloning site in which the passenger domain site is deleted from autotransporter proteins of bacteria. A vector contains TEV protease cleavage site inserted to the cloning site. The autotransporter protein has an amino acid sequence of sequence number 1. The passenger domain site has a sequence of 29th to 785th residue in an amino acid of sequence number 1.
