Provided are a reagent and method for assaying the total protein S activity in a sample, which are capable of assaying the activity of all proteins S present in a sample, that is, assaying total protein S activity. According to the present invention, a reagent for assaying total protein S activity in a sample includes activated proteins C, a surfactant, phospholipids, calcium ions, activated coagulation factor V, activated coagulation factor X, and prothrombin and thrombin substrates. According to the present invention, a method for assaying total protein S activity in a sample is characterized in that a surfactant is present during a reaction to assay the total protein S activity.
