A method for detection of pathogenes in pileate fungi (Basidiomicetes) such that while preparing a preparation from mycelium a layer thereof of 2- is mixed with a phosphotungstic acid (PTA) at pH 5.5-6.5 and the prepared mixture contracts the prepared mixture with exposure of 150-180 seconds, coated onto an grid-substrate, dried, and a photograph is taken by an electronic microscope; assessing by external characters is carried out on the basis of images (photographs) of the contrasted mycelium preparation; and while preparing the fungal fruiting body preparation a sample is taken from a pileus of fruiting body shaped as a triangle with a side of 2.5- with the further contrasting treatment in PTA at pH 5.5-6.5 and exposure of 90-180 seconds, applying the mixture onto the grid-substrate and drying. The final assessment of the contrasted preparation of the fungal fruiting body by external characters is carried out on the basis of images (photographs). Such structure-morphological objects as bacteria, microscopic fungi and viruses are detected as a whole in the same preparation and for a short period of time. In addition bacteria, phages thereof (viruses), microscopic fungi and fragments thereof (organelles) in preparations of mycelium (fruiting body) of pileate fungi (Basidiomicetes) are detected and fixed on images with magnification by 10000-25000 times, and viruses of rod, bacillary and isometric form are detected and photographs thereof are taken at magnification by 25000-45000 times. |
