A lipid- and membrane-free, catalytically-active, heteromeric, mammalian cytochrome P450Âcytochrome-P450 reductase complex is disclosed comprising a cytochrome P450 (CYP450) and a cytochrome P450 reductase (POR) polypeptide, each adapted to be immobilised onto a lipid- and/or membrane-free surface, wherein the CYP450 and POR polypeptides do not form a CYP450-POR chimaeric protein and wherein the CYP450 catalytic activity of the complex is POR-dependent. The CYP450 may be CYP3A4. The CYP450 and POR polypeptides may comprise a biotinylation motif such as acetyl co-enzyme A carboxylase biotin carboxyl carrier protein (BCCP). A method of assessment and/or prediction of drug metabolism with the use of the complex is also described, along with kits or beads comprising one or more lipid- and membrane-free, catalytically-active, heteromeric, mammalian CYP450-POR polypeptide complexes. |
