PURPOSE: A method for preparing mutant E.coli having an inducible promoter of chb operon or asc operon with a constitutive promoter is provided to prevent yield reduction. CONSTITUTION: A method for preparing mutant E.coli using cellobiose comprises a step of inducing mutation in wild type E.coli by substituting chb operon inducible promoter with constitutive promoter, substituting asc operon inducible promoter with the constitutive promoter, or chb operon inducible promoter and asc operon inducible promoter with the constitutive promoter. The wild type E.coli is E.coli MG1655. The constitutive promoter has a base of sequence number 2.
