Provided is a composition or the like containing lipid particles packed with double-stranded nucleic acid molecules, the composition being a polynucleotide 17 to 30 nucleotides long which has an antisense strand comprising a nucleotide sequence complementary to a consecutive 17-nucleotide sequence of a target gene mRNA, and which has a sense strand comprising a nucleotide sequence complementary to the 1st to 17th nucleotide toward the 3' terminus from the 5' terminus of the antisense strand, where saccharides for binding to a nucleotide at a specific location of the antisense strand and the sense strand are, each at a specific ratio, a deoxyribose, or a ribose in which the hydroxyl group at position 2' has been substituted with a modifying group. The lipid particles are lipid particles having a lipid bilayer membrane, a constituent component of which is a lipid derivative of a water-soluble substance, a fatty acid derivative, or a derivative of an aliphatic hydrocarbon.
