PROBLEM TO BE SOLVED: To provide a method of detecting K-ras gene mutations, wherein blood can be used as inspection material in place of cancer tissue.

SOLUTION: The method of detecting K-ras gene mutations includes a process for extracting circulating DNA from a blood sample that is selected from the group consisting of a subject's blood, plasma, and serum and a process for reacting the extracted circulating DNA with a PNA probe containing a forward primer, a reverse primer, and the codon 12 and the codon 13 of K-ras to perform a PCR reaction to detect whether the codon 12 and the codon 13 of K-ras have mutated.
