摘要 <P>PROBLEM TO BE SOLVED: To provide a method for engineering an immunoglobulin containing at least one modification in a structural loop region of the immunoglobulin and determining the binding of the immunoglobulin to an epitope of an antigen, wherein the unmodified immunoglobulin does not significantly bind to the epitope. <P>SOLUTION: The method includes steps of: providing a nucleic acid encoding an immunoglobulin containing at least one structural loop region; modifying at least one nucleotide residue of at least one of the structural loop regions; transferring the modified nucleic acid in an expression system; expressing the modified immunoglobulin; contacting the expressed modified immunoglobulin with an epitope; and determining whether or not the modified immunoglobulin binds to the epitope.
