PURPOSE: A trans-splicing group I which specifically recognizes single nucleotide mutant transcript is provided to specifically treat and diagnose cancer cells expressing mutant transcript. CONSTITUTION: A trans-splicing group I ribozyme targets and specifically recognizes Kra G12V RNA. The ribozyme has 'GCAGCU' or 'GACAGC' as an inner nucleotide sequence. The ribozymes is AS100KRib1 of sequence number 25, AS300KRib1, of sequence number 26, AS100KRib2 of sequence number 27, or AS300KRib2 of sequence number 28. The trans-splicing group I ribozyme contains a reporter gene at 3'-exon. The reporter gene is a gene encoding chloramphenicol acetyl transferase, Renila luciferase, red fluorescence protein, green fluorescence protein, secreted placental alkaline phosphatase, or Herpes simplex virus-thymidine kinase. |
