PURPOSE: A preparation method of opuntia humifusa pills is provided to enable users to easily intake all kinds of nutrients from opuntia humifusa without the loss of the nutrients. CONSTITUTION: A preparation method of opuntia humifusa pills comprises the following steps: extracting an opuntia humifusa mixture using an alcohol water solution, and concentrating the same to obtain opuntia humifusa mucilage; drying and crushing extraction residue obtained after extracting stems, fruits, and roots of opuntia humifusa; mixing 100 parts by weight of residue powder, 3-15 parts by weight of opuntia humifusa flower powder, and 5-30 parts by weight of opuntia humifusa mucilage; and adding 5-20 parts by weight of fermented wheat dough to the mixture before molding the mixture into pills. The wheat dough is obtained by mixing flour and water, and fermenting the mixture at 25-40 deg. C. for 2-4 days.
