PURPOSE: A method for measuring sex hormone in saliva is provided to detect trace amounts of estradiol and to quickly and objectively determine menstrual regulation. CONSTITUTION: A method for measuring estradiol concentration comprises: a step of treating strong base of pH 12-14 to collected saliva; a step of heating at 70-90°C and cooling; a step of removing digestive enzymes and bacteria from saliva by centrifuging; a step of neutralizing; and a step of removing mucosaccharide-proteins. The saliva is collected one hour after getting up. The strong base includes NaOH, LiOH, KOH or Ca(OH)_2.
