PURPOSE: A method for purifying saliva by denaturing proteins is provided to enhance accuracy and to detect trace amount of hormone. CONSTITUTION: A method for purifying saliva comprises: a step of treating strong base of pH 12-14 to collected saliva; a step of heating and cooling the saliva; a step of neutralizing the saliva; and a step of removing mucosaccharide-proteins from the saliva. The method is a process of pretreating the saliva for quantitation of steroid hormone such as sex hormone or adrenal cortical hormone. The strong base includes NaOH, LiOH, KOH, or Ca(OH)_2. The removal of mucosaccharide-proteins is treating mucosaccharide-proteins enzyme. The enzyme is used for decomposing Nacetylhexosamine-[1→4]glycosidic bond.
