<P>PROBLEM TO BE SOLVED: To provide a recombinant antigen-binding region and an antibody and a functional fragment containing such an antigen-binding region that are specific for CD38, also to provide a nucleic acid sequence encoding the antibody, a vector containing the sequence, a pharmaceutical composition and a kit with an instruction for use, and further to provide an isolated novel epitope of CD38 and a method of use therefor. <P>SOLUTION: The invention provides an isolated antibody or a functional antibody fragment that contains an antigen-binding region that is specific for an epitope of CD38, where the antibody or the functional fragment thereof is able to mediate killing of a CD38+ target cell (LP-1 (DSMZ: ACC41) and RPMI-8226 (ATCC: CCL-155)) by antibody-dependent cellular cytotoxicity ("ADCC") with an at least two- to five-fold better efficacy than the chimeric OKT10 antibody having SEQ ID NOS: 23 and 24, when a human PBMC cell is employed as an effector cell, and when the ratio of effector cells to target cells is between about 30:1 and about 50:1. <P>COPYRIGHT: (C)2012,JPO&INPIT |
