CHIMPANZEE ADENOVIRUS VACCINE CARRIER

PROBLEM TO BE SOLVED: To provide adenoviral vaccine carriers applicable to human hosts which can be replicated in high-level in human complementation cell lines, is highly immunogenic, devoid of neutralizing B cell epitopes that cross-react with the common serotypes of human adenoviruses, complying with the safety RCA standards promulgated by regulatory agencies, and can be used in priming/boosting that are suitable for use in humans.

SOLUTION: The present invention provides recombinant replication-defective adenoviral vectors derived from chimpanzee adenoviruses and methods for generating recombinant adenoviruses in human E1-expressing cell lines. The invention provides compositions and methods suitable for use for the delivery and expression of transgenes encoding immunogens against which a boosted immune response is desired. The invention provides methods for generating clinical grade vector stocks suitable for use in humans. The invention contemplates the use of vectors comprising transgenes which encode tumor-associated antigens in vaccines and pharmaceutical compositions for the prevention and treatment of cancer.
