The semi-methylation linker has a first strand and a second strand, wherein the cytosines in the first strand are all modified by methylation, none of the cytosines in the second strand are modified by methylation, and the 3'-end sequence of the first strand is complementary to the 5'-end sequence of the second strand.</p>
