<P>PROBLEM TO BE SOLVED: To provide a method for synthesizing a nucleic acid, with which a reaction can be conducted in a fluid (flow) and improved coupling efficiency is brought about, by using a highly dispersible liquid phase support. <P>SOLUTION: In the method for synthesizing an oligonucleotide, a hydrophobic group-linked nucleoside of formula (1) [in the formula, R<SP POS="POST">1</SP>is a 1C-12C alkylene group, R<SP POS="POST">2</SP>is a 1C-22C alkylene group, each of R<SP POS="POST">3</SP>and R<SP POS="POST">4</SP>is independently a 1C-22C alkyl group and so on, R<SP POS="POST">5</SP>is a single bond or a 1C-22C alkylene group, each of R<SP POS="POST">6</SP>is independently a 6C-30C alkyl group, n is 2-6, X is a hydrogen atom, a hydroxyl group, and so on, and Z is an adenyl group, a guanyl group, and so on, the polar group of which is optionally protected] or an oligonucleotide and a nucleoside phosphoroamidite dissolved in a nonpolar solvent are brought into contact with an acid-azole complex compound or a solution thereof. <P>COPYRIGHT: (C)2012,JPO&INPIT |
