A method and an apparatus for making a transport frame and processing a transport frame capable of flexible packet design are provided to use data transmission space due to classifying the packet without additional data such as padding data and to design the packet flexibly. One or more packet transmitted through a transfer frame is received(210). An initial location information about a first packet is created(220). The first packet is the initial packet including the beginning of packet and end of packet of the received packet within the transfer frame. The transfer frame creates the transfer frame including at least one of the packet and the initial location information.
