The invention relates to a nucleic acid encoding a fusion protein comprising a conditional retention domain consisting of an FRB domain followed by a retention motif in the endoplasmic reticulum. The expression vectors and cells transformed by such vectors are also part of the present invention. In another embodiment, the invention also comprises methods for conditional retention of a protein of interest in the endoplasmic reticulum using these vectors, and to reagent kits comprising such nucleic acids or vectors.
