摘要 PURPOSE: A method for preparing linear alpha-(1,4)-glucan is provided to maintain enzyme activity and to improved thermal stability. CONSTITUTION: An immobilized enzyme is prepared by immobilizing amylosucrase to a pH-sensitive polymer carrier. The amylosucrase is derived from Neisseria polysaccharea, Deinococcus geothermalis or Alteromonas macleodii. The pH-sensitive polymer carrier is a methacrylic acid-methylmethacrylate copolymers(eudragit L100 and S100), methacrylic acid-methylacrylate copolymer(MPM-05), or methacrylic acid-methylacrylate-methyl methacrylate copolymers, or methyl methacrylate copolymers(MPM-06). A method for preparing linear alpha-1,4-glucan comprises: a step of adding immobilized enzyme to perform enzyme reaction; a step of centrifuging the enzyme reactant to separate precipitate; and a step of freeze-drying the precipitate.
