发明名称 METHOD FOR PRODUCING PERIPHERAL BLOOD MONOCYTE-DERIVED PLURIPOTENT STEM CELLS
摘要 Induced pluripotent stem cells having properties similar to ES cells and the same genomic information as the genomic information of a patient are produced from human peripheral blood monocytes without leaving, within the cells, the genes used for producing the iPS cells. Initialization genes of the cells are mounted on a Sendai virus vector that is capable of sustained expression and that does not exert the activity of integrating foreign genetic information into chromosomes. After introducing and expressing the aforementioned genes into peripheral blood monocytes, the vector genome RNAs including the aforementioned genes are removed from the cells, thereby establishing induced pluripotent stem cells. The abovementioned Sendai virus vector is capable of efficiently producing, in an extremely simple manner, induced pluripotent stem cells having the same genomic information as the individual providing the differentiated cells. Moreover, the Sendai virus vector is also safe because the risk of tumor formation is low, and may become an important tool for safely and efficiently providing cell replacement therapy by providing useful induced pluripotent stem cells.
