Microscope and microscopy method

摘要

The microscope (M) has a microscope lens provided for focusing an illumination beam into a sample, a detection optical path and a demodulation unit, where the illumination beam is phase-modulated with modulation frequency in a partial area along a cross-section. A polarization-changing element i.e. electric-optical modulator (EOM), is provided in an illumination beam path that is arranged downstream of a phase plate (P) i.e. spatial light modulator, where the phase plate comprises two regions with different phase influences and designed in a rotatable manner. An independent claim is also included for a method for operating a laser scanning microscope.
