<P>PROBLEM TO BE SOLVED: To provide a novel, simple, quick and practical method for detecting RNAs in a sample. <P>SOLUTION: The method for detecting RNAs in a sample containing organism-derived impurities includes: (1) a step for producing a circularized single-stranded DNA probe by circularizing a single-stranded DNA with complementary sequence in a target RNA; (2) a step for hybridizing the circularized single-stranded DNA probe obtained in the step (1) with RNA in the sample; (3) a step for synthesizing a single-stranded DNA from the RNA-DNA hybrid obtained in the step (2) by a rolling circle amplification (RCA) method with the RNA as a primer; (4) a step for detecting the target RNA by adding a marker to be combined with the target RNA synthesized in the step (3) and detecting the marker; and further, as an option, (5) a step for forming a nick only in the RNA strand of the RNA-DNA hybrid with RNase H after the step (2). <P>COPYRIGHT: (C)2012,JPO&INPIT |
