<P>PROBLEM TO BE SOLVED: To provide an ergo-thionase useful in the specific quantity determination of ergothioneine, a gene which encodes the same, a transformant containing the gene, a method of producing the ergo-thionase, a quantitative procedure of ergothioneine using the ergo-thionase. <P>SOLUTION: A Burkholderia sp. HME 13 strain which has ergo-thionase productivity is found out, the ergo-thionase is refined from the strain, and has become clear to act on L-ergothioneine specifically. Moreover, the cloning of the gene which encodes the enzyme is carried out, the high expression is carried out by Escherichia coli, thereby it has become possible to carry out the extensive and easy preparing of the enzyme.
