Protein misfolding cyclic amplification of prion protein performed in the presence of a polysaccharide sulfate

A method for efficiently amplifying abnormal prion protein (PrPSc) derived from bovine spongiform encephalopathy (BSE). Based on protein misfolding cyclic amplification (PMCA). Normal prion protein (PrPC) is used as a source and PrPSc derived from BSE is used as a seed. The method comprises stirring, mixing, incubating, and sonicating both the PrPC and the PrPSc repeatedly in the presence of a polysaccharide sulphate. In particular the polysaccharide sulphate can be dextran sulphate or pentosan sulphate, wherein the dextran sulphate can be dextran sulphate sodium or dextran sulphate potassium.
