An illuminating light source (22) such as a laser light source illuminates a sample (36) with illuminating light (24), and a scanner (28) deflects the illuminating light in such a way that the illuminating light optically scans the first side of sample and outputs a detection light (38). A main beam divider (26) splits the illuminating light from detection light. A detector assembly (44) detects the lateral limited detection light. An illuminating light source (50) illuminates second side of sample with illuminating light (46) and outputs the detection light (51). An independent claim is included for method of operating microscope.
