Isolation and/or purification of definite amount of nucleic acid from a nucleic acid containing sample, comprises: (a) contacting the nucleic acid containing sample with a defined amount of a nucleic acid binding phase, where the amount of nucleic acids in the sample exceeds the binding capacity of the amount of used nucleic acid binding phase; (b) binding the nucleic acids to the nucleic acid binding phase at a pH value, which is below the pKa value of at least one of the protonizable groups; and (c) eluting the nucleic acids at a pH value above the binding pH value. Isolation and/or purification of definite amount of nucleic acid from a nucleic acid containing sample, comprises: (a) contacting the nucleic acid containing sample with a defined amount of a nucleic acid binding phase with following properties in which (i) the nucleic acid binding phase comprises nucleic acid binding ligand that comprises at least one protonizable group, (ii) the nucleic acid binding ligands are present bonded to a carrier, and (iii) the nucleic acid binding phase comprises a surface having a low charge density, where the amount of nucleic acids in the sample exceeds the binding capacity of the amount of used nucleic acid binding phase; (b) binding the nucleic acids to the nucleic acid binding phase at a pH value (binding pH value), which is below the pKa value of at least one of the protonizable groups; and (c) eluting the nucleic acids at a pH value above the binding pH value, where a defined amount of nucleic acid is obtained. An independent claim is included for a kit, for isolation and/or purification of definite amount of nucleic acid from a nucleic acid containing sample, comprising the nucleic acid binding phase. |
