RPM1 (BCDIN3D) is identified as a novel class of RNA methyltransferase that is capable of methylating an exposed 5' monophosphate of nucleic acids, particularly RNAs such as microRNAs including miR-145. Compositions for methylating substrate nucleic acids are provided that comprise RPM1. Also, methods of methylating substrate nucleic acids are described as are methods for screening RPM1 as a drug target, and methods for diagnosing cancer in a biological sample.
