Incorporation of DNA into higher plant genome - by specified recombinant DNA techniques

Incorporation of a DNA sequence (I) into the genome of a higher plant is effected by : (a) isolating a complete gene, eg coding for a seed protein, and determining its coding and regulatory DNA sequences by sequence analysis; (b) inserting the gene into a plasmid vector and excising the coding sequences without significantly altering the regulatory sequences; (c) re-ligating the regulatory sequences and simultaneously creating a unique restriction site (URS) in the active region of the regulatory sequences;
