PURPOSE: A method for making an image of sodium/calcium activity using two-proton fluorescent probe is provided to selectively and easily load a complex together with an calcium ion. CONSTITUTION: A two-proton fluorescent probe is denoted by chemical formula 1. In chemical formula 1, X is O, S or NH. The probe forms a complex by reacting with calcium ions and contains a compound of chemical formula 1. A method for preparing the probe comprises: a step of refluxing a mixture of 6-bromo-N-methyl-2-naphthylamine, proton-sponge, and tert-butyl bromoacetate to prepare compound B of chemical formula 2; a step of stirring a mixture of the compound B, benzoxazole, Pd(II)OAc, PPh3, CuI, and CsCO_3 to prepare compound A of chemical formula 3; and a step of mixing the compound A, 1-hydroxybenzotriazole, and compound D of chemical formula 4.
