<P>PROBLEM TO BE SOLVED: To provide a technique by which, when microparticles labeled with a plurality of fluorescent dyes are to be multi-color measured with a plurality of optical detectors, fluorescence intensity from each fluorescent dye is accurately calculated and presented to a user. <P>SOLUTION: A fluorescence intensity correcting method includes procedures of receiving, with optical detectors differing in optical wavelength band, fluorescence generated from fluorescent dyes excited by irradiating microparticles multiple-labeled with a plurality of fluorescent dyes whose fluorescent wavelength bands overlap one another, and approximating, with the linear sum of monochromic-dyed spectra obtained with microparticles individually labeled with a plurality of fluorescent dyes, a measured spectrum obtained by collecting detection values from the optical detectors. In the procedures, the approximation of the measured spectrum with the linear sum of the monochromic-dyed spectra is performed using a constrained least squares method. <P>COPYRIGHT: (C)2012,JPO&INPIT |
