<P>PROBLEM TO BE SOLVED: To provide a homogeneous method for determining enzymatic activity of an analyte in a sample in in-vitro diagnosis to enable simultaneous determination of the amount of enzyme in the same reaction mixture and determination of enzyme activators or inhibitors. <P>SOLUTION: Provided are (a) a process for preparing and incubating a reaction mixture comprising a sample, at least one substrate capable of being bound or transformed with an objective enzyme to be measured, and a first component and a second component in a signal forming system, wherein the first component and the second component in the signal forming system interact so as to generate a detectable signal when they are mutually brought to be spatially close and, at this time, the first component in the signal forming system is bound with or brought to be bound with the substrate during incubation and the objective enzyme to be measured is brought to be bound with the second component of the signal forming system during the incubation; and (b) a process for measuring the activity of the enzyme in the sample with a process for measuring the signal in relation to the enzymatic activity in the sample. <P>COPYRIGHT: (C)2012,JPO&INPIT |
