Disclosed is an oligonucleotide for targeted alteration of a duplex DNA sequence. The duplex DNA sequence containing a first DNA sequence and a second DNA sequence which is the complement of the first DNA sequence. The oligonucleotide comprises a domain that is capable of hybridising to the first DNA sequence and which comprises at least one mismatch with respect to the first DNA sequence and also comprises at least one section that contains at least two modified nucleotides having a higher binding affinity compared to naturally occurring A, C, T or G nucleotides, wherein - at least one modified nucleotide is a LNA that is positioned at a distance of at least one nucleotide from the at least one mismatch and wherein, optionally, the oligonucleotide contains at most about 50% LNA modified nucleotides; and - at least one modified nucleotide is a C7-propyne purine or a C5-propyne pyrimidine.
