TRANSGENIC NON-HUMAN ANIMAL CAPABLE OF PRODUCING HETEROLOGOUS ANTIBODY

PROBLEM TO BE SOLVED: To provide transgenic non-human animals capable of producing heterologous antibodies and methods for producing human sequence antibodies which bind to human antigens with substantial affinity.

SOLUTION: A cell including at least one artificial gene encoding at least a portion of an immunoglobulin polypeptide produces a detectable amount of immunoglobulin binding a predefined human antigen. The immunoglobulin polypeptide has a sequence substantially the same as an immunoglobulin polypeptide secreted from a hybridoma obtained from a transgenic mouse which contains a homozygous pair of functionally disrupted endogenous heavy chain alleles, a homozygous pair of functionally disrupted endogenous light chain alleles, at least one copy of a human immunoglobulin light chain transgene, and at least one copy of a human immunoglobulin heavy chain transgene. The immunoglobulin binds to the human antigen at an affinity constant (Ka) of at least 1.1×109M-1.
