The invention provides a magnetic fluorescence kit for rapid detection of microorganisms and its preparation and usage. The kit includes immunomagnetic micro beads and immunofluorescent micro beads, which specifically bind microorganisms of interest. The preparation comprises the preparation of immunomagnetic micro beads and immunofluorescent micro beads. The usage includes: a sample, immunomagnetic micro beads and immunofluorescent micro beads are added to a buffer; the microorganisms have antigenic determinants which can simultaneously bind immunomagnetic micro beads and immunofluorescent micro beads; microorganisms that bind to immunomagnetic micro beads can be enriched by magnetic separation; and qualitative or quantitative judgments are made by measuring the fluorescence intensity.
