According to one embodiment of the present invention, a total internal reflection fluorescence imaging device comprises: a metal nanostructure layer including a metal thin film and a nanostructure formed on the metal thin film; a light source unit for providing incident light which is totally internally reflected from the metal nanostructure so as to produce an evanescent wave localized in a horizontal direction between the metal nanostructure and a specimen disposed on the metal nanostructure; and a fluorescence image extraction unit which extracts a fluorescence signal generated from the specimen by means of the evanescent wave located in the horizontal direction, and obtaining an image from the extracted fluorescence signal.
