PURPOSE: A method for preparing 2,5-diketopyperazine compounds is provided to simplify preparation process and to ensure eco-friendly process. CONSTITUTION: A method for preparing 2,5-diketopyperazine compounds of chemical formula 1 comprises a step of reacting linear dipepeptide compounds of chemical formula 4 under the presence of water solvent at 80°C-180°C. The compound of chemical formula 1 is a stereoisomer with an optical activity. In the method, used reaction solvent is ethanol, methanol, tetrahydrofurane, 4-dioxane, N,N-dimethylformamide, or dimethyl sulfoxide.
