The present invention relates to a composition for the treatment or prevention of bone disease comprising a glyceollin as an active ingredient. The glyceollin constituting the active ingredient of the present invention promotes breakdown of the Runx2 protein in osteoblasts, reduces the Rankl mRNA level and so suppresses the differentiation of osteoclasts, and has greatly alleviated the signs of osteoporosis in an animal model of osteoporosis induced by the administration of a large dose of vitamin D. The composition of the present invention has very high potential to be developed as a drug or food for the prevention or treatment of, inter alia, osteoporosis and bone disease caused by bone metastasis of cancer cells.
