PROBLEM TO BE SOLVED: To provide an SSR primer pair for identifying cultivars of Lolium perenne L. and Lolium x boucheanum Kunth simply, quicky, and accurately, a specific marker gene, and a method for cultivar identification.SOLUTION: The primer pair is designed by using base sequences on both sides of an SSR region on a genome of Lolium perenne L. and Lolium x boucheanum Kunth. The base sequence of a specific marker of a gene of an identification object is obtained by bulking genes of Lolium perenne L. and Lolium x boucheanum Kunth to be objects to eliminate a variation of genes between individuals and specifically amplifying a cultivar-specific SSR by PCR. The method for cultivar identification includes detecting a difference in length between amplified gene fragments. The method for identifying cultivars of Lolium perenne L. and Lolium x boucheanum Kunth has a fewer number of necessary steps in comparison with a conventional variety differentiation method based on morphological characteristics, heading date, or the like, and uses bulk DNAs of a plurality of individuals as a template DNA. |
