<P>PROBLEM TO BE SOLVED: To provide a molecular sensor capable of highly accurately quantifying an intracellular concentration of inositol 1,4,5-triphosphate (IP<SB>3</SB>). <P>SOLUTION: This molecular sensor is composed of a ligand binding part of an IP<SB>3</SB>receptor and two kinds of fluorescent substances inducing fluorescence resonance energy transfer (FRET). An ECFP (enhanced cyan fluorescent protein) or its modified product and an EYFP (enhanced yellow fluorescent protein) or its modified product are preferably used as the two kinds of the fluorescent substances. When a gene which encodes the molecular sensor is introduced into a cell, etc., and then the cell (SH-SY5Y cell) which has expressed the molecular sensor is bound to the IP<SB>3</SB>, an efficiency of the FRET between the two kinds of the fluorescent substances (e.g., the ECFP and the EYFP) is changed, so that the IP<SB>3</SB>is measured by sensing a fluorescence intensity ratio. <P>COPYRIGHT: (C)2005,JPO&NCIPI |
