PROBLEM TO BE SOLVED: To provide a method for identifying a target base sequence by effectively reacting a double stranded sample nucleic acid and a probe nucleic acid forming a double strand without a thermal denaturation process.SOLUTION: The method for identifying the target base sequence comprises a step of recognition in which the target base sequence in a base sequence of double stranded sample nucleic acid is identified by allowing the sample nucleic acid and a nucleic acid probe partially forming a double strand to be present in a reaction solution and measuring the degree of chain-substitution reaction. The nucleic acid probe forming a double strand comprises the first probe which is single stranded nucleic acid consisting of a base sequence complementary to the target base sequence and the second probe which is single stranded nucleic acid consisting of a base sequence complementary to the first probe, and the ratio of the base length of the second probe to the base length of the first probe ([the base length of the second probe]/[the base length of the first probe]) is ≥0.5 and <1. The temperature of the reaction solution is <65°C. |
