PROBLEM TO BE SOLVED: To provide a granulocyte colony stimulating factor (G-CSF) analog, a composition containing the analog and related compositions.SOLUTION: There is provided a method for producing modified G-CSF polypeptide, wherein a second chemical moiety is indirectly bonded through a first chemical moiety and amino acid sequence comprising a specific sequence is comprised. The method includes the following steps: (a) bonding the first chemical moiety to an outer-loop in vitro; and (b) indirectly bonding the second chemical moiety to the outer-loop through the first chemical moiety. In the method, the first chemical moiety is polysaccharide, the second chemical moiety is polyethylene glycol, N-terminal methionine of amino acid sequence comprising a specific sequence exists or not exists, and the biological activity in which the modified G-CSF is bonded to a G-CSF receptor is kept. |
