PROBLEM TO BE SOLVED: To provide an antibody enzyme eliminating the function of a chemokine receptor CCR which is a coreceptor of an AIDS virus by degrading the chemokine receptor CCR5, and utilized for preventing the infection with the AIDS virus or treating the AIDS; and to provide an anti-HIV medicament utilizing the antibody enzyme.
SOLUTION: A monoclonal antibody is produced by using a peptide constituting the N-terminal region of the chemokine receptor CCR5 as an immunogen, and amino acid sequences and base sequences of variable regions of the heavy chain and the light chain of the monoclonal antibody are determined. The three-dimensional structure is estimated and the presence or absence of a triadic structure of catalyst residues is confirmed by carrying out molecular modeling as the amino acid sequence. Finally, it is found that the variable region of the light chain of the monoclonal antibody (5A2, 6A2, 2B8) having the triadic structure of the catalyst residues functions as the antibody enzyme.
COPYRIGHT: (C)2007,JPO&INPIT
|
