摘要 <p>PURPOSE: A method for typing H3N2/H1N1 type of influenza A virus and detecting amantadine resistant mutant is provided to ensure high accuracy and sensitivity. CONSTITUTION: A method for typing H3N2 or H1N1 type of influenza A virus and detecting amantadine resistant mutant using single base extension comprises: a step of isolating genome RNA of influenza A virus and preparing cDNA; a step of amplifying PCR of M2 protein coding base sequence; and a step of performing single base extension the amplified product with primers of sequence numbers 5 and 6 in a reaction solution; and a step of analyzing fluorescence pattern. The reaction solution contains dideoxyadenosine triphosphate(ddATP), dideoxycytosine triphosphate(ddCTP), dideoxyguanosine triphosphate(ddGTP), and dideoxythymidine triphosphate(ddTTP) labeled with fluorescence materials with different colors.</p>
