<p>The invention relates to a method for genotyping blood samples for red blood cell and/or platelet surface antigens containing antigens of ‰¥99% prevalence and antigens of <99% prevalence in the population, the method comprising the steps of:
(a) extracting DNA from a blood sample,
(b) subjecting the DNA to multiplex PCR, targeting at least three red blood and/or platelet surface antigen genotypes in each reaction mix of at least two reaction mixes,
(c) subjecting amplification products to analysis by electrophoresis, and
(d) comparing the electrophoresis bands obtained in step (c) with bands of positive controls of the red blood cell and/or platelet surface antigens, with identical bands indicating the presence of the red blood cell and/or platelet surface antigens in the sample,
wherein in each PCR reaction mix targets of antigens of ‰¥99% and <99% prevalence are combined such that alternating bands of antigens of ‰¥99% prevalence and of <99% prevalence are obtained in step (c). The method is cost-efficient and allows easy interpretation of results.</p> |
