The present invention addresses the need to improve the yields of virus from plasmid transient transfection when grown in cell culture systems. In particular, it has been demonstrated that transfection and suspension cell culture systems provide for high titer viral production. In other embodiments, the inventors have shown that there is improved recombinant viral production with cells grown in serum-free conditions, and in particular in serum-free suspension culture and then transfected by static or attached to cell stacks, or suspension transfection with the addition of selective additives.
