Sulphates of estrone, equilenin, equilin, b -estradiol, a - and b -dihydroequilenin are prepared from pregnant mares' urine by preparing a concentrate which is dissolved in an aqueous medium and salted out with an inorganic salt, e.g. sodium and potassium salts of sulphuric, phosphoric or hydrochloric acid.ALSO:Estrogen compositions which may be administered orally, topically or by intra-venous injection, are prepared from pregnant mares' urine by preparing a concentrate which is dissolved in an aqueous medium and salting out the solution with an inorganic salt, removing the precipitate and separating the estrogens from inorganic salt therein. Specified inorganic salts are the alkali metal, preferably sodium and potassium, salts of sulphuric, phosphoric or hydrochloric acids. The precipitate may be extracted with an aliphatic ketone, such as acetone or methyl ethyl ketone, containing a small amount of an alcohol, such as methanol, ethanol, isopropanol or butanol. Finally water may be added to the solution and the product concentrated so as to remove solvent leaving a concentrated aqueous solution. Buffers (many specified) may be added to maintain pH 6.5-7.5. Specified carriers and drying agents are dextrose, galactose, pectin, gelatine, dextran, polyvinylpyrrolidone and preferably lactose. Specified estrogens present are the sulphates of estrone, equilin, equilenin, b -estradiol, a - and b -dihydroequilenin.
