The present invention relates to a method for producing a biosensor for an in vitro screening system for identifying anti-infective substances, comprising the steps of a) cloning random genomic DNA fragments from at least one organism of interest upstream of a promoter-less reporter gene into a vector, in order to generate a random genomic library, b) transforming said random genomic library into a host strain, c) selecting clones with anti-infective-responsive reporter expression from said host strains, comprising treating said clones with either i) at least one known anti-infective substance, or ii) at least one potentially anti-infective substance, and iii) comparing said treated clones from i) or ii) with at least one untreated control sample, and d) identifying those clones exhibiting a different anti-infective-responsive reporter expression as biosensors. The invention furthermore relates to an in vitro screening system for identifying anti-infective substances as produced according to the invention, preferably comprising at least two complimentary biosensor clones. The invention also relates to uses of said screening system, in particular for high-throughput screening (HTS) in order to identify molecules which inhibit bacterial growth, and can be used as antimicrobially effective substances. |
