PURPOSE: A method for preparing a protoplast relating to cassava is provided to obtain a regenerative protoplasts and novel cassava starch. CONSTITUTION: A method for preparing cassava starch comprises: a step of preparing friable embryonic callus from an explant of cassava; a step of isolating protoplast from the embryonic callus; a step of introducing a vector having an antisense gene of gbss gene to suppress amylose production; a step of producing an embryo by the protoplast; and a step of regenerating plant from the protoplast and isolating from a tuber.
