PURPOSE: A two-photon fluorescence probe is provided to ensure strong fluorescence by reaction with sodium cation and to enable image of sodium cation distribution in cells or tissues. CONSTITUTION: A two-photon fluorescence probe contains 2-acetyl-6-(dimethylamino)naphthalene as a reporter and 1,7-diaza-15-crown-5 as a sodium cation receptor. The probe is denoted by chemical formula 1. The probe forms a complex by binding metal ions. A probe is used for imaging distribution of sodium cation in cells or tissue. The tissue is nerve conduction tissue, muscle, or heart. The cells are mouse hippocampus cells, or astrocytes. A method for manufacturing the probe comprises: a step of preparing 6-acryl-2-[N-methyl-N-(carboxymethyl)amino]naphthalene(compound A) and N-2(-methoxyphenyl)-N'(4-amino-2-methoxyphenyl)-1,7-diaza-15-crown-5(compound B); a step of mixing the compound A, 1-hydroxybenzotrazole, and 1,3-dicyclohexyl carbodimide in dichloromethane to prepare a mixture C; a step of adding compound B to the mixture C to prepare a mixture D; a step of removing an organic layer to prepare a product E; and a step of purifying product E.
