A vaccine is produced by attentuating canine hepatitis virus by a number of serial passages in trypsinized tissue culture of dog kidney, uterus or testicle, followed by propagation in a trypsinized pig kidney tissue culture in a medium which supports growth and/or maintenance of pig kidney cells, for example: 8 parts Earles solution, 1 part 5% lactalbumin hydrolysate and 1 part inactivated horse serum; 8 parts Earles-Simms solution, 1 part 5% lactalbumin hydrolysate and 1 part inactivated horse serum, or # 199 of Morgan, Morton and Parker supplemented with 10% inactivated horse serum. The virus containing liquid is harvested and may be used as a vaccine or be freeze-dried to give a solid which may be diluted with distilled water for use as a vaccine.
|
